• W2364372428 abstract "OBJECTIVE:To investigate the different reaction of lung adenocarcinoma lung A549 and lung adenocystic carcinoma cells A2 treated by all-trans retinoic acid, and manifest the derivation of the differene and find the method of drug treatment against lung adenocarcinoma. METHODS: A549 and A2 cells were cultured and treated by all-trans retinoic acid. The flow cytometry was used to detect the percentage of cells at debris, apoptosis, G0/G1 phase, S phase, G2/M phase, diploid and aneuploid cells. The Western blot method was used to detect the expressions of caspase3 and p21. The giemsa staining method was used to observe the morphologic change of cells. The immunohistochemical method was used to detect the expression of antioncogene Rb. The electronmicroscope was used to observe the change of mitochondria. RESULTS: Compared with the control group, the proportions of apoptosis cells and necrosis cells were increased. The proportions of lipl-oid cells in A549 and A2 were increased, from (94.79±4.50)%, (94.24±3.12)% to (98.64±0.16)% and (97.75±1.95)%, respectively. The expression of caspase3 was up-regulated in A2(from 3 320±168 to 7 353±204) and p21(from 4 336±256 to 6 171±188) in A549. The dividing cells, asymmetrical karyokinesis and pantomorphia cells were reduced in A549, while the volume of cells was augmented, and the multinucleatedgiant cells were increased in A2. The expression of Rb showed up-contrast to that of the control groups in lung cancer cell lines A549 and A2 by immunohistochemical method. The mitochondria changed into swell and round appearance, and the crista miochondriales got unclear, which were more significant in A2.CONCLUSIONS: The cell cycle, genetic expression, appearance, and structure of mitochondria of cells A549 and A2 can be changed by all-trans retinoic acid. Some differences of reactivation to the same drug in cells A549 and A2 are probably concerned with the structure of mitochondria. The induction differentiation therapy after operation is going to increase the therapeutic effects and elevate the quality of living patients who attacked by adenocarcinoma of lung." @default.
