- W2365323920 abstract "Objectives To explore the effect of interleukin-17A(IL-17A) on myocardial fibrosis in mice with viral myocarditis(VMC).Methods Wild-type(WT) and IL-17A-deficient(IL-17A-/-) mice on the BALb/c background were intraperitoneally injected with coxsackie-virus B3(CVB3) to establish VMC models.Cardiac tissues were obtained 0 d,14 d,28 d and 42 d after CVB3 injection and made into paraffin sections.Myocardial histopathologic changes were observed by hematoxylin eosin and Masson staining.Furthermore,expressions of matrix metalloproteinase-2(MMP-2),matrix metalloproteinase-9(MMP-9) and tissue inhibitors of metalloproteinase-1(TIMP-1) in cardiac tissues were quantified by SP immunohistochemical method.Results Compared with WT mice,IL-17A-/-mice developed significantly less severe myocardial inflammation and fibrosis in VMC and survival rate was improved.Additionally,cardiac expressions of MMP-2 and MMP-9 markedly reduced in IL-17A-/-mice at 14 d and 28 d [14 d subgroup: 0.21±0.02 vs.0.32±0.04,0.22±0.02 vs.0.38±0.05,all P0.05;28 d subgroup: 0.14±0.02 vs.0.22±0.02,0.15 ±0.01 vs.0.26 ±0.03,all P 0.05],whereas TIMP-1 in IL-17A-/-mice significantly increased at 14 d(P0.05).Ratios of MMP-2 / TIMP-1 and MMP-9 / TIMP-1 in WT group firstly increased,then decreased,and the peak appeared at about 14 d.While ratio of MMP-2 / TIMP-1 and MMP-9 / TIMP-1 in 14 d,28 d and 42 d subgroups of IL-17A-/-group significantly decreased compared with those in 0 d subgroup(P0.05),but there were no significant differences among the ratios of the 3 subgroups(P0.05).Conclusions Genetic deletion of IL-17A contributes to the balance of MMP-2 / TIMP-1 and MMP-9 / TIMP-1 system.IL-17A may mediate the imbalance of collagen degradation and synthesis system in murine VMC,which may indicate that inhibiting IL-17A contributs to myocardial fibrosis.Targeting IL-17A is an attractive therapeutic option for myocardial fibrosis in the development of VMC." @default.
