- W2365841389 abstract "Objective To explore the significance of the differentially expressed genes in mouse bone marrow after exposure to neutron irradiation.Methods Sixty male BALB/c mice were randomly divided into control group and irradiation group(30 each).Mice in irradiation group were exposed to whole body irradiation with fission neutron reactor for an absorption dosage of 3.0Gy.The animals were sacrificed at 6,24 and 72 hours after irradiation.The basic pathological changes in the mouse bone marrow after irradiation were observed by HE staining.Whole genome expression oligonucleotide microarray was performed 6 hours after irradiation to observe the changes in the whole genome in murine bone marrow.A part of the differentially expressed genes were further identified by RT-PCR.Results Hematopoietic cells in murine bone marrow of irradiation group were progressively reduced after neutron irradiation with an absorption dosage of 3.0Gy.Massive simulating blood pool was found in the bone marrow of the irradiation group mice,with only a few hematopoietic cells existing at 72 hours after irradiation.Six hours after 3.0Gy neutron irradiation,there were 390 differentially expressed genes in the murine bone marrow,among them 195 genes were up-regulated and 195 genes were down-regulated.The functions of the differentially expressed genes were related to metabolism,growth and development,cell signaling pathway,stress response,proliferation and death,immune response,cell connections,material transport,cell cycle,and some genes with unknown function.The gene function was possibly associated to bone marrow injury induced by neutron irradiation,including cell cycle regulation(ccng1,ccne2),cell proliferation and apoptosis(bax,bbc3,traf5) and cytokine synthesis(psg18,zbtb32,irf4,t2bp,etc.).Conclusion Hundreds of differentially expressed genes may appear in murine bone marrow after neutron irradiation,among which ccng1,bax,bbc3,ccne2,traf5 and irf4 may be the sensitive genes of bone marrow injury induced by neutron irradiation." @default.
