- W2366202005 abstract "Objective To investigtate the effects of adenovirus mediated e NOS gene transfer on monocrotaline-induced pulmonary hypertension and pulmonary vascular remodelingin rats.Methods Twenty-four male RD rats were randomly divided into control( C)group,Monocrotaline( M) group,Monocrotaline-Lac Z( M-Lac Z) group and Monocrotaline-e NOS( M-e NOS) group with 6 rats in each group.600 μl of Ad CMVe NOS titered 5 × 10~9 PFU / ml was injected intracracheally into rat lung from M-e NOS group.Rats from MLac Z group received equal dose of Ad CMVLac Z,rats from C and M group received equal dose of normal saline.Three days later,pulmonary hypertension model from M,M-Lac Z and M-e NOS group were establised by intraperitoneally injecting 60 mg / kg of monocrotaline( diluted to 1ml using normal saline).Rats from C group received equal dose of normal saline.Three weeks later,mean systematic arterial pressure( MSAP),mean pulmonary arterial pressure( MPAP),heart rate( HR),pulmonary vascular remodeling( percentage of muscularized arteries in small pulmonary vessels) and right ventricular hypertophy index( ratio of right ventricle to left ventricle +septum weight) were measured.e NOS immunoblot,nitric oxide( NO) and interleukin 6( IL-6) level in lung s were also measured.Results The MPAP,pulmonary vascular remodeling degree,right ventricular hypertophy index and IL-6 level in M-e NOS group were significantly lower than those in M and M-Lac Z group,higher than those in C group( all P 0.05).The e NOS protein and NO level in C group were significantly higher than those from M and M-Lac Z group,significantly lower than those from M-e NOS group( P 0.05).There were no significant difference with regard to MSAP and HR in rats in all group( P 0.05).Conclusion Adenovirus mediated e NOS gene transfer can upregulate e NOS expression,elevate NO level and decrease IL-6 release in lung,which lead to selective pulmonary vessel dilation and anti-inflammation effect and alleviates monocrotaline-induced pulmonary hypertension and pulmonary vascular remodelingin rats." @default.
