- W2366437368 abstract "Objective To evaluate the inhibitory effect and toxicity of juglone administered via different routes on the nude mice model bearing subcutaneously transplanted tumor.Methods Human hepatocellular carcinoma BEL-7402 cells were subcutaneously injected into nude mice.The resultant tumors were taken and prepared into small tissue blocks,which were implanted subcutaneously into nude mice to establish mouse models bearing human hepatoma.Juglone was administered at different doses by intraperitoneal injection and local injection.The general condition of the tumor-bearing mice and the tumor growth were observed for pharmacodynamic study of the quinone.Results The inhibitory effect of 600 μg/kg,300 μg/kg and 150 μg/kg juglone on the tumor growth in nude mice treated was studied.The results revealed that these doses of juglone had no significant impact on tumor growth,but the NK cell activity decreased in nude mice treated treated with 600 μg/kg and 300 μg/kg juglone.Compared with the positive control group(5-Fu),NK cell activity of nude mice treated with 600 μg/kg juglone was not significantly different(P0.05),while the NK cell activity of nude mice treated with 300 μg/kg and 150 μg/kg juglone showed a significant difference(P0.05,P 0.01).The tumor-bearing nude mouse models,which received locally injected 4.5 mg/kg,3 mg/kg and 1.5 mg/kg juglone,showed that juglone effectively inhibited the tumor growth,with an inhibition rate of 78.24%,66.57% and 48.94%,respectively.Juglone at doses of 4.5 mg/kg and 3 mg/kg had the same anti-tumor effect when compared with the positive control group(P0.05,P0.05),but there were significantly reduced subcutaneous fat,weight loss,and showed death in the 4.5 mg/kg juglone group.In the tumor-bearing mice,which received juglone at pH 7.4 and pH 4.0 administered in a dose of 600 μg/kg and 150 μg/kg,respectively,the tumor growth was effectively inhibited,similar to that in the tumor-bearing mice of positive control group(5-Fu)(P0.05),while the difference of anti-tumor effects of 150 μg/kg juglone at different pH was not significant.The anti-tumor effects of juglone at the same concentration but different pH showed no significant difference(P0.05),but there were less liver metastases in the pH 4.0 group.Conclusions Juglone administered via different routes inhibits the growth of transplanted human liver cancer BEL-7402 in nude mice.However,the drug shows a narrow therapeutic range and with certain side effects." @default.
