- W2366666083 abstract "Objective To evaluate the inactivation effect of Thiazole Orange(TO) photochemical method on PRV and Sindbis virus in plasma.Methods The absorption spectrum of TO in plasma were measured and PRV and Sindbis viruses were used as indicator viruses.PRV and Sindbis virus were each added to TO-plasma,to prepare TO-virus-plasmas.Viral inactivation kinetics was studied first,and the optimal condition for viral inactivation was determined by orthogonal test,in which 3 factors,namely TO concentration(C),irradiation time(T),light intensity(I),and each at 3 levels were studied.At last,the viral inactivation effects of TO photochemical method were tested on uncovered samples,on samples placed in plastic bags,and on oxygenated samples in bags under the optimal condition.Results The absorption peak of TO in plasma was at 476 nm.By irradiation with 1.06E-01 w·m-2·nm-1 light,most viruses in 60 μmol/L TO-virus-plasma were inactivated after 5 min,then the inactivation effect entered a plateau phase.Orthogonal test results indicated that the optimal inactivation condition for both PRV and Sindbis virus was I=1.33E-01w·m-2·nm-1,T=20 min,C=80 μmol/L.Under this condition,≥6.13 LogTCID50(naked samples,n=8),(5.66±0.29) LogTCID50(samples in bags,n=4) and(6.32±0.55) LogTCID50(oxygenated samples,n=4) PRV were inactivated,(5.86±0.29) LogTCID50(naked samples,n=8),(3.88±0.25) LogTCID50(samples in bags,n=4) and(6.00±0.35) LogTCID50(oxygenated samples,n=4) Sindbis virus were inactivated.Conclusion TO photochemical method is an effective method for virus inactivation,and the effect is enhanced by increasing the oxygen content of the blood." @default.
