- W2366793750 abstract "AT-1420 is methylphenylalanine mustard. Experimental studies showed that AT-1420 at the doses of 0.155-1mg/kg/d×7 can induce 81-100% inhibition of W-256 in rats, with 50-100% regression of the tumors. This therapeutic effect was similar to that of Nitrocaphan (AT-1258) under the doses of 3-3.5mg/kg/d×7. AT-1420 showed inhibitory effect also on the growth of S-180, Hepatoma and L2, giving 48-50%, 28% and 41-50% inhibition rates, respectively, The prolongation of life span of mice bearing EAC was 177% in the AT-1420 treated group as compared to the control. However, AT-1420 showed no significant inhibitory effect on the growth of S-37. In the experiments with W-256 in rats and L2, S-180 in mice, oral administration of AT-1420 (1.5-2mg/kg/d) showed similar effect as that with intraperitoneal injection (0.6mg/ kg/d).AT-1420 significantly inhibited tne incorporation of 3H-TdR into EAC cells.The LD50 of AT-1420 for rats was determined to be 5.4 (4.39-6.63) mg/kg by one intraperitoneal injection or l.23mg/kg injected intraperitoneally daily for 7 consecutive days. The LD50 of AT-1420 for mice was 8(5.28-10.2) mg/kg by one ip injection or 1.12mg/kg injected i.p. daily for 7 days.The data showed that AT-1420 possesses high antitumor activity, wide antitumor spectrum. Its therapeutic dose is smaller than that of AT-1258. So AT-1420 appeared to be a promising anticancer drug, it is worthwhile to study further the possibility of its clinical use." @default.
