- W2367236052 abstract "Objective:In the project, the condition to induce DCs from cord blood, normal human peripheral blood and chronic myeloid leukemia (CML) cells and their anti leukemia response were investigated.Methods:The mononuclear cells were cultured with the cytokines (GM CSF+IL 4+TNF α) for inducing DC. The morphologic features were analyzed with optical microscopy and the phenotype of DC was detected by FACS with CD 1a , CD 80 , CD 83 and HLA DR monoclonal antibodies. DCs were pulsed by the CML cell lysates and incubated with autologous T lymphocytes for 5d to induce antigen specific CTL. Cytotoxicity assay was measured by MTT method.Results:It was proved that the DCs derived from the different sources showed a typical morphology of DC. A high expression of phenotypes was found in CB DC, PB DC and CML DC stimulated by the cytokines. The antigen loaded DCs from different sources, all of them have strong capacity of stimulating CTL reaction.Conclusion:It is concluded that cord blood, normal human and CML patients peripheral blood cells could successfully induce functional DCs; They have the ability of inducing CTL reactionto clear leukemia cells. It is a novel idea that expanding DC in vitro and enhancing their antitumor immunefunction and DC based tumor vaccines may be used as an efficient immune therapy for leukemia and others." @default.
