- W2367395762 abstract "[Objective]To study the effect of total flavonoids of Astragalus(TFA)on expression of PPARγand FXR in hepatic cirrhosis model induced by DMN in rats.[Methods]Fiftythree male SD rats were allocated randomly into Control group(10rats)and model group(43rats),rats in model group were given DMN by intraperitoneal injection for 4weeks.At the 3rd experimental week the model rats were randomly divided into model group(14rats)and TFA intervention group(14rats in low-dose group,15rats in highdose group),rats in intervention group were given TFA at different dosage correspondingly for 4 weeks.At the end of theexperiment rats were sacrificed,liver tissue collected,keptin formalin,paraffin embedded,4μm slices prepared,dewaxed,washed,microwave antigen retrieval and 5% BSA blocked.Then slices were incubated with the first antibodies(PPARγantibody concentration:160;FXR antibody concentration:150)at 37°C,using PBS instead of primary antibody as negative control.SABC method was adopted by using twostep immunohistochemical detection kit.[Results]Compared with the normal group,PPARγand FXR expression in model group were significantly reduced.Compared with the model group, PPARγand FXR expressions in TFA intervention groups were increased significantly,particularly in the highdose group.[Conclusion]TFA have anti-fibrotic effect through upregulating the expression of PPARγand FXR in DMN hepatic cirrhosis model." @default.
