• W2367736929 abstract "Objective:To observe the protective effect of and valsartan (a Angiotensin Receptor Blocker,ARB) on the kidney function of diabetic rats, to study their possible mechanisms and discover the new method for treating diabetic nephropathy. Methods: 47 adult male SD rats were divided randomly into five groups ; normal control group ( CON ) , diabetic control group ( DM ) , diabetic group treated orally with Valsartan ( ARB) , diabetic group orally treated with YISHENGTANG 1 (YS1), diabetic group orally treated with YISHENTANG 2 (YS2). At the end of 12 weeks, 24-hour urine were collected. The rats were weighted, then sacrificed without anesthesia. The parameters of biochemistry and hormone were analysed, and the average thickness of glomerular basement membrane was measured determined by electron microscope the image analyzer. Results; Diabetes was successfully induced by injection of STZ in all DM groups. The body and liver weight in diabetes rats were significantly lighter than those of the CON, and the body weight in every group of ARB, YS1, YS2 was significantly greater than that of DM. The liver weight of YS2 was significantly greater than that of DM. The renal relative weight in all diabetes rats was significantly greater than that of CON, ARB was significantly lighter than DM. Plasma glucose and HbAlc in all the diabetes rats were significantly greater than that of CON, and all of treated groups were significantly decrease than that of DM. Plasma creatinine concentration in YS1 and YS2 were significantly less than that of the other three groups, and in YS1 was significantly less than that of YS2. 24 hours urine volume and urinary albumin content in every group of diabetes rats was significantly greater than that of CON, in every group of treated groups was significantly less than that of DM. 24 hours urine total protein in diabetes rats were significantly greater than that of CON. ET-1 content in 24 hours urine and renal tissue was significantly greater in every group of diabetes rats than in CON, and significantly less in every of treated diabetes rats than in DM. There were no markedly difference about plasma ET concentration in every group of the five ones. The average thickness of glomerular basement membrane was significantly greater in every group of diabetes rats than that in CON, and significantly less in every group of the treated diabetes rats than in DM ones. The content of 24 hours urinary albumin was significantly correlative with ET-1 level of 24 hours urinary and renal tissue, and no markedly correlative with plasma ET concentration. 24 hours urine volume was greater in every group of the diabetes rats than CON ones. Conclusion; YISHENTANG decreased the urine albumin excretory rate and the concentration of plasma glucose and creatinine, and lowered the ET content in renal and urine, also postponed the glomerular basement membrane thickening in diabetes rats. YISHENTANG exhibited the similar effects with ARB for preventing and treating diabetic nephropathy. It was probably that the effects of YISHENTANG on preventing and treating diabetic nephropathy would attribute, in parts to decreasing ET formation in kidney, interfering the effect of ET, and ameliorating the glucose metabolism." @default.
