• W2368278006 abstract "Objective To identify the relationship between the four systemic lupus erythemstosus (SLE)-related genes and the pathogenesis of SLE,provide information for SLE at molecule level.Methods Four genes,which expressed significantly different between SLE patients and healthy controls,were found by gene chips' screening.Chromosome location found that they were located at SLE linkage chromosome zone. TaqMan real time quantitative PCR was used for detecting KLRC1,TRIM,EGR1 and IL-4R mRNA expression in 55 SLE patients and 30 controls.Results In the peripheral blood mononuclear cells of SLE patients,ΔCt of gene TRIM was significantly lower than healthy controls(P＜0.05),the gene expression level was 0.61 times of that of normal controls,ΔCt of genes IL-4R and EGR1 were significantly higher than those of healthy con- trols(P＜0.05),the gene expression level was 2.62 and 2.03 times of those of normal controls respectively,ΔCt of gene KLRC1 was significantly higher than that in healthy controls(P＜0.01),the gene expression level was 5.33 times of normal controls.In the peripheral blood T cells of SLE patients',ΔCt of gene TRIM was sig- nificantly lower than that of healthy controls(P＜0.01),the gene expression level was 0.56 times of that of nor- mal controls;ΔCt of genes IL-4R and EGR1 were significantly higher than those of healthy controls(P＜0.05),the gene expression level was 2.55 and 1.74 times of those of normal controls respectively,ΔCt of gene KLRC1 was significantly higher than that of healthy controls(P＜0.01),the gene expression level was 5.19 times of that of normal controls.Because the ratio of peripheral blood B lymphocytes in lymphocytes was so low,gene detection data was instable so statistical analysis was not carried out.There was no correlation be- tween the four SLE-related genes and SLEDAI score.Conclusions The expression of TRIM,IL-4R,EGR1 and KLRC1 genes in SLE patients are significantly different from those of healthy controls,but has no rela- tionship with the activity of the disease." @default.
