- W2368373218 abstract "Objective:To study the effects of Dengzhanhua Injection on proliferation of cultured mesangial cells(MC) induced by endothelin- 1(ET-1). Methods: The proliferation of MC induced by ET- 1 and followed by incubation with Dengzhanhua Injection was measured by MTT method and flow cytometry(FCM). Results: (1)After incubation for 48h, ET - 1 stimulated the proliferation of MC with a dose dependent manner (P0.05 or P0.01 ). But Dengzhanhua Injection (45,90,180,360 ug/ml) together cultured with10-8 mol/L ET-1 for 48h, can inhibit the proliferation of MC with dose dependent manner(P0.05 or P0.01).(2) ET-1(10~(-9)～10~(-7)mmol/L) incubated for 24 h, then DNA was measured and cell cycle analyzed by FCM, We found ET - 1 increased percentage of cells in S phase, and decreased that of G_0/G_1 phase (P0.05) also with a dose dependent manner. While Dengzhanhua Injection (45,90,180,360 ug/ml) together with 10~(-8) mol/L ET- 1 incubated for 24 h, the percentage of cells in Q_0/G_1 phase was increased,and S phase reduced with a dose dependent manner.Conclusion:Dengzhanhua Injection can inhibit DNA synthesis of MC induced by ET- 1, delay cell cycle, suppress MC proliferation, so that it can alleviate glomerular sclerosis and its progression, which could be one of the mechanisms of Dengzhanhua Injection's treating renal disease." @default.
