- W2368686148 abstract "Objective: To study the effects of valsartan and angiotensin Ⅱ(AⅡ) on nitric oxide synthase (NOS) activity and nitric oxide (NO) level in cultured human coronary artery endothelial cells in the state of hypoxia. Methods: The cultured cells of the third generation were divided into control group, AⅡ (10 μmol/L) group, valsartan (10 μmol/L) group and AⅡ(10 μmol/L) with different concentrations of valsartan (0.1 μmol/L, 1 μmol/L and 10 μmol/L) groups. Valsartan and AⅡ were added when the cultured endothelial cells had reached 60 percent of culture bottle. Both of NOS activity and NO level were examined after the incubation for 32 h or in state of hypoxia for 4 h. Results: The NOS activity and NO level were lower in AⅡ group than those of control group(P 0.01). There were no significant differences in NOS activity and NO level between valsartan and control group (P 0.05). The NOS activity and NO level increased in AⅡ with different concentrations of valsartan groups compared to those of AⅡ group in state of hypoxia. The effects of valsartan were positively related to the concentrations(P 0.01). Conclusion:Angiotensin Ⅱcould decrease NOS activity and NO level in human coronary artery endothelial cells and the effect could be reversed by valsartan in state of hypoxia. The result suggested that valsartan could improve the function of human coronary artery endothelial cells and prevent or reverse atherosclerosis in state of hypoxia." @default.
