- W2368895174 abstract "Objective To explore the activation and differentiation ofγδT cells after being stimulated with influenza virus and analyze its cytotoxic effects.Methods Peripheral blood mononuclear cells(PBMCs) from 6 donors were stimulated in vitro with H1N1 、 H3N2and B group of influenza viruses for 12-24hours;then the activation surface marks(CD25and CD69) and the differentiation marks(CD4and CD8) were detected by flow cytometry.γδT cells were isolated by cytometry using anti-γδantibodies.Lactate dehydrogenase(LDH) assay was used to measure the cytotoxic activity ofγδT cells.Isolated PBMCs were stimulated with recombinant human granulocyte-macrophage colony-stimulating factor(rhGM-CSF) to obtain differentiated monocyte-derived macrophages(MDMs).Results After stimulating with different influenza virus strains,the activation markers CD25and CD69in isolatedγδT cells were significantly higher than that in the uninfected group(CD25 : 58.2%,50.8%,49.5%and 19.9%in H1N1,H3N2,influ B-infected and uninfectedγδT cells,respectively;CD69 : 61.4%,47.0%,30.5% and 11.1%in H1N1,H3N2,influ B-infected and uninfected γδT cells,respectively).After simulation,γδT cell tended to be both CD4and CD8positive(CD4 + CD8 + : 33.8%,29.7%,29.1% and 14.4%in H1N1,H3N2,influ B-infected and uninfectedγδT cells).LDH assay indicated that the cytotoxic effects of stimulatedγδ T cells on H1N1-and H3N2-infected MDMs(35.46%-52.17%,28.46%-41.34%,respectively) were significantly higher than that in influ B-infected group(18.94%-25.98%).Conclusions γδT cells showed quick response to different influenza viruses and differentiated into CD4 + CD8 + T cells that demonstrated cytotoxic activity in the influenza virus-infected MDM model." @default.
