- W2369184400 abstract "Determining bacteria cell population for gaseous hydrocarbon-oxidating bacteria(GHOB) is necessary in microbiological prospecting of oil and gas(MPOG) and the main determining methods used at present,the most probable number(MPN) test and the plate test,are time-consuming and troublesome.A simple and convenient culture flask test is recommended instead.The flasks contain one or another gaseous hydrocarbon(as only carbon sources) and proper nitrogen and phosphorus sources in cultures as well as a special indicator for the growth of GHOB bacteria.The soil samples under test are converted by dispersing them in distilled water into aqueous samples and the laters are diluted successively,step-by-step,and cultivated at 30℃for 14 days.The change of a limpid aqueous sample to turbid one or the appearing of a layer of bacteria on the surface of sample with its color changed from red to yellow denote the growth of GHOB bacteria.The bacterial cell population is determined by the same procedures as in common flask tests.By using GHOB culture flasks of type JH manufactured in Yangtze University,the bacterial cell population of GHOB in lots of soil samples is determined and some results obtained(in cell/g) are as follows: 2.0×10~6 for MOB(averaged over 3 parallel tests),6.0×10~6 for EOB(averaged over 2 parallel tests),1.3×10~4 for POB(averaged over 2 parallel tests),2.5×10~3 for BOB (averaged over 2 parallel tests),and 1.5×10~5 for P/BOB(averaged over 3 parallel tests),being similar to that by MPN method,2.0×10~6,6.0×10~6,1.3×10~4,3.0×10~3,and 1.6×10~5 and that by plate method,1.9×10~6,6.3×10~6,1.1×10~4,3.2×10~3,and 1.8×10~5 for the above mentioned soil samples,respectively." @default.
