- W2369684219 abstract "Objective To develop a real-time fluorescence quantitative PCR method for analyzing the levels of VEGF mRNA from various organs of health adult rats. Methods A pair of primers and TaqMan probe which spanned two exons were designed for the gene. The PCR product was cloned to plasmid pGEM-T by TA ligetion to get the standard template. Then,evaluation of performance of the PCR which consisted of specificity,sensitivity and repeatability were established. Total RNAs from normal various tissues (lung,heart,liver,kidney,brain,stomach,intestine and submaxillary gland) of 10 rats were extracted to be quantified for the levels of VEGF gene. Results DNA sequencing and agarose gel analysis of PCR products confirmed the high specificity of the method. CV within group was 2.0% ～6.7% and between groups was 3.5% ～10.6%. The correlation coefficient(r) was more than 0.99. VEGF mRNA level in lung was the highest(P0.05),while differences between other tissues were not significant. Conclusion The real-time quantitative PCR for detecting VEGF gene was established successfully with the character of high specificity,sensitivity and repeatability. The method and the analysis of levels of VEGF gene from various organs of health adult rats laid a good foundation for studying in depth levels of VEGF gene of unhealthy rats." @default.
