- W2369702563 abstract "AIM To study effects of quercetin(Que) on prevention of cardiac hypertrophy in rats induced by isoprenaline(Iso) and its mechanism. METHODS Cardiac hypertrophic models of rats were induced by a continued administration of isoprenaline(Iso) 0 02 mg·kg -1 sc twice a day for six weeks, the ratio of heart wet weight and left heart wet weight to body weight were significantly increased.The myocardial lipid peroxidation(LPO) contents increased, superoxide dismutase (SOD) activities decreased, the calcium contents of myocardium and aorta artery were also increased. Intracelular free calcium concertration [Ca 2+ ] i was measured with Ca 2+ sensitive fluorescent indicator Fura 2/AM in cultured neonatal rat heart cell. RESULTS By a continued administration of Iso for sex weeks, the ra tio of heart wet weight and left heart wet weight to body weight significantly increased, the myocardial LPO contents incresed, SOD activities decreased the calcium contents of myocardium and aorta artery also incresed. After treated with quercetin(Que) 75, 150 mg·kg -1 and verpamil 10 mg·kg -1 for five weeks, the cardiac hypertrophy significantly improved. The myocardial LPO contents decreased, SOD activities increased, the calcium contents of myocardium and aorta artery were also decreased. Intracellular free calcium concentration [Ca 2+ ] i was measured with Ca 2+ sensitive fluorescent indicator Fura 2/AM in cultured neonatal rat heart cells. There were no effects of Que on the action in normol cells while Que 1～100 μmol·L -1 might inhibit [Ca 2+ ] i elevation induced by Iso and H 2O 2 in dose dependent. CONCLUSION Cardiac hypertrophy and improving cardiovascular disease might relate to scavenging oxygen free radicals and decreasing the [Ca 2+ ] i of the myocardial cells." @default.
