- W2369870173 abstract "Objective To investigate the effect of ACEI enalapril on the activity of p38MAP kinase in the obstructed kidney after unilateral ureteral obstruction(UUO) of rats. Methods The UUO models were induced by ligating the left ureter. Rats were divided into normal control (sham operation),model and treatment groups (enalapril 10 mg·kg-1·d-1 by gastric gavage from 24 h before the obstruction to day 28 after the induction of UUO). Rats were sacrificed at 1 h, 3 h, 6 h, 12 h, 1 d, 3 d, 5 d, 7 d, 14 d, 21 d and 28 d after UUO was initiated. p38MAPK activity was assayed by immunohistochemical staining and specific substrate phosphorylation with immunoprecipitation and Western blotting. TGF β1 protein and mRNA expression was analyzed with immunohistochemical and in situ hybridization. Results A basic p38MAP kinase activity(A,0.22±0.06)was detectable in normal kidney. p38MAP kinase pathway was rapidly activated at 1 h(0.45±0.14,P 0.01) and the activity was steadily increased by 12 h(0.91±0.07,P 0.01) after UUO induced, and then declined gradually. However, the activity of p38MAP kinase was evident again at 3 d, peaked at 7 d(0.93±0.06, P 0.01), and then decreased. A basic expression of TGF β1(A,4.32±1.72) was detectable in normal kidney. Significant upregulation of TGF β1 was tested at 3 d(13.55±6.33,P 0.01) and this was steadily increased by 7 d(26.78±8.77,P 0.01). Interestingly, blockade of AngⅡwith enalapril significantly suppressed the activation of p38MAPK(reduced by 36%～65%,P 0.01). Similarly, inhibition of AngⅡresulted in obvious suppression of TGF β1 expression in obstructed kidney(reduced by 33%～46%,P 0.01). Conclusion Enalapril significantly alleviates renal tubulointerstitial fibrosis by markedly suppressing the expression of TGF β1 in obstructed kidney,which may partly relate to the inhibition of p38MAP kinase signaling pathway." @default.
