- W2370398816 abstract "Objective:To study the differentiation and DNA ploid patterns of cells that had stem cell markers during the cancerous process of cervical epithelium and investigate its significance and prognostic value. Methods:Ninty eight cervix samples were divided into 4 groups,20 cases of normal squamous epithelium(NSE),19 cases of dysplasia squamous epithelial hyperplasia(DSEH),39 cases of squamous carcinoma in situ(SCIS),and 20 cases of invasive squamous epithelial carcinoma(ISEC) of cervix. The expression of keratin 19 and β1 integrin was detected by immunohistochemical SP method respectively and Feulgen staining was carried out simultaneously.Different morphological parameters,DNA mass, and DNA ploid in cells, which co-expressed Keratin 19 and β1 integrin, were detected by using HMIAS-2000 image analysis system. Results:The cells with co-expression of Keratin 19 and β1 integrin only located at the basal layer of epithelium in NSE.During the cancerous process of cervix, the number of Keratin 19 andβ1 integrin co-positive cases increased with positive cells moving upwards from the basal layer.Different cell morphological parameters,DNA mass, and the number of DNA aneuploidy increased. The difference was significant between different groups(P0.05) indicating the abnormal proliferation and terminal differentiation disorder of the cells that had stem cell markers. Conclusion:During the cancerous process of cervical epithelium, the neoplastic cells that have stem cell markers with the abnormal proliferation and differentiation disorder may be the main cellular clone to initiate tumor formation and maintain the unlimited proliferation of tumor cells. The obvious difference in the rate of DNA aneuploidy in different cases of primary neoplasia may be one of the reasons for the dualism of the biological features of primary neoplasia. The existence of diploid tumor cells may be related with the relatively stable growth features of some cases of primary neoplasia in a long period." @default.
