- W2370487639 abstract "OBJECTIVE To develop a novel non-iral gene delivery systems biopolymer-PEI-DNA polyplexes and investigate their transfection efficiencies in vtiro. METHODS The GPDa and GPDb were prepared as follows by first mixing the plasmid DNA and cationic polyethlenimine PEI together,then the resulted complexes were incubated for 10 minutes at room temperature,followed by addition of cationic gelatin and anionic gelatin . The surface charge of GPD a and b were measured by photon correlation spectroscopy. Estimation of transfection efficiency was measured by luciferase activity in C 3 cells and Hela cells. RESULTS The zeta potential of GPDa polyplexes increased with increased amount of gelatin,but the zeta potential of GPDb polyplexes decreased.The GPDa polyplexes showed about 15%～20% increased transfection efficiency compared to positive control in C 3 cells and Hela cells.The transfection efficiency of GPDb in C 3 cells increased with increased amount of gelatin,but transfection efficiency in Hela cells decreased. CONCLUSION The GPDb could be selective for C 3 cells." @default.
