- W2370623983 abstract "Objective To study the lowest effective dosage and mechanism of atorvastatin on atherosclerosis. Methods The atherosclerotic animal model was established in Wistar rats by feeding cholesterol-rich diet.Then these atherosclerotic rats were treated with atorvastatin 2.5mg/kg·d,5mg/kg·d and 10mg/kg·d respectively for six weeks.Thereafter the levels of lipids were detected and the pathologic changes of rat aortas tissue were ob- served by means of hematoxylin-eosin stain(HE staining).The expressions of inflammatory factors mRNA in the aorta tissue of the rats were analyzed using RT-PCR.Results The levels of lipids in rats with 5mg/kg·d,10mg/ kg·d atorvastatin intervention were different from those of atherosclerotic model rats(P0.05),but the levels of lipids in 2.5mg/kg·d atorvastatin interveneing rats had no difference with those of atherosclerotic model rats(P0.05).The pathology of aorta tissue showed that early phase plaques occurred in atherosclerotic model rats and only the arterial intimae were thickening with vessel smooth muscle cell hyperplasia in all atorvastatin interveneing rats,but no plaque occurred.The expressions of ICAM-1,MMP-2,TIMP-1 mRNA in atherosclerotic model rats were increased significantly compared with those of normal rats(P0.01)and the expressions of inflammatory factors mRNA in all intervened rats were decreased compared with atherosclerotic rats(P0.01).Conclusion Atorvastatin effects on atherosclerosis through lipid-lowering and anti-inflammation;the latter is independent of lipid-lowering.The lowest effective dosages of atorvastatin in anti-inflammation and lipid-lowering are 2.5mg/kg·d,5mg/kg·d respectively." @default.
