This nucleoprotein structure presents a sizable barrier to the process of RNA transcdption, inhibiting both the accessibility of the general transcrip- tion machinery to promoter sequences and the binding of upstream regulatory pmteinsL How do transcription factors contend with nucleosomes that mask their recognition sequences? A recent convergence of biochemistry and gen- etics has identified a conserved muiti- subunit complex, the SWi-SNF complex, that uses the energy of ATP hydrolysis to drive transcription factors onto nu- cleosoma| transcription-factor-binding sites. The huge size of the SWI-SNF complex (2 MDa, about half the size of a ribosome) suggests that it is truly a pro- tein machine that plays several roles in chromatin remodeling and transcrip- tion-factor function. Currently, the SWI-SNF complex is the only well- characterized factor known to facilitate transcription by remodeling chromatin. Recently however, two groups have Jescribed the biochemical identifi- cation of Drosophila factors that also appear to mediate an AT[dependent disruption of chromatin ~,3.
