- W2370736700 abstract "Objective To study the metabolic kinetics of trans-resveratrol and the effects of selective UGT inhibitors on the metabolism of trans-resveratrol in human liver microsomal enzyme.Methods The metabolisms of trans-resveratrol concentration in the incubation pool was determined by UFLC-MS method.The Michaelis-Menten parameters Km and Vm in human liver microsomes were initially estimated by analyzing Origin 7.5.The clearance(CLint)was calculated for in vitro enzyme to Trans-resveratrol.Curcumin and quercetin as selective UGT inhibitors were used to investigate their inhibitory effects on the metabolism of Trans-resveratrol.Results Two metabolites,4'-O-monoglucuronide resveratrol(M-1) and 3-O-monoglucuronide resveratrol(M-2),were identified by liquid chromatography/mass spectrometry.The Vm and Km of M-1 were(1.08±0.06) nmol/min/mgand(192.05±30.46) μmol/L;(2.20±0.10) nmol/min/mgand 34.82±6.95μmol/L of M-2.The formation of M-2 is much faster than M-1.The metabolism of resveratral was significantly inhibited by Curcumin and quercetin.Conclusion The formation of M-2 contribute to the clearance of resveratrol in human liver microsomes.Curcumin and quercetin as the selective UGT inhibitors have obvious inhibition on metabolism of resveratrol in vitro and will give some direction for the research of the interaction of resveratrol." @default.
