• W2371427350 abstract "Objective To investigate the effect of cordycepin alone or combined with chemotherapy on the proliferation,migration and apoptosis induction of colon cancer cells in vitro.Methods Effect of cordycepin on the proliferation and migration of colon cancer cells:serum pharmacological methods were used to facilitate the study.A total of 20 BALB/c mice was randomized into two groups and fed with diet supplemented with cordycepin or saline respectively.All mice were sacrificed on day 3 and their serums were harvested respectively.After the serums which included effective component were collected,the effect of cordycepin alone or combined with chemotherapy on colon cancer cell line SW1116 was evaluated.SW1116 cells were divided into various groups(cordycepin group,chemotherapy group,cordycepin+chemotherapy group and saline group as a control) and cultured in medium mixed with cordycepin,chemotherapy,cordycepin+chemotherapy,respectively.cells treated with normal medium alone were used as blank control group.Cell viability and proliferation was determined by WST-8 assay using cell counting kit-8(CCK-8).Apoptosis was assessed by Hoechst 33258 fluorescent microscopy,transmission electron microscope and Annexin-V/PI flow cytometry.Results(1)Proliferation inhibition assay:CCK-8 assay indicated that proliferation of SW1116 cells were inhibited after various treatments for 96 h and 120 h.Proliferation inhibiting rate of cordycepin+cisplatin group was significantly higher than those of other groups(P=0.02).(2)Cell migration assay:cell migration of cordycepin+cisplatin group was 38.17±4.41 which was significantly lower than other groups(F=91.04,P0.05).(3)Cell apoptosis detection: cell apoptosis detection using Hoechst 33258 fluorescent microscopy showed that large amount of apoptotic cells(brilliant blue staining) were induced after treatments with cordycepin+cisplatin.Quantitation of apoptosis using annexin V and PI flow cytometry indicated the apoptotic rate of cordycepin+cisplatin group [(28.10±4.21)%] was significantly higher than those of other groups[Cordycepin group(16.19±5.65)%,Chemotherapy group(14.90±3.56)%,blank control group(6.83±1.65)%,P0.05].There were no significant differences among the cordycepin polysaccharide group,cordycepin group,chemotherapy group,and blank control group as compared with each other.Conclusions(1)Cisplatin and cordycepin have significant anti-proliferative,anti-migration,and pro-apoptosis effect on human colon cancer cell line SW1116 cells.The combination of cisplatin and cordycepin can promote the anti-tumor effect,which suggests that cordycepin may has adjuvant effect for chemotherapy;(2)Study based on nude mouse peritoneal carcinomatosis model of human colon cancer cells suggests that cordycepin and cordycepin can significantly inhibit the growth of peritoneal dissemination nodules.The combination of cisplatin and cordycepin can promote the anti-tumor effect." @default.
