- W2371433698 abstract "Background and purpose:Dendritic cells (DC) are a unique population of antigen-presenting cells (APC) and also one of the most potent initiators and modulators of the immune response.Gene transfer vector is the important ingredient in cancer gene therapy.The aim of this study was to transfect dendritic cells originating from CD34+ hematopoietic stem cells with human AFP and SLC,and to observe the immune response against hepatocellular carcinoma.Methods:CD34+ hematopoietic stem cells were harvested after mobilization by chemotherapy and G-CSF.CD34+ hematopoietic stem cells were induced to differentiate between dendritic cells by cytokine IL-4,GM-CSF or TNF-α.DCs transfected with Lenti-AFP/Lenti-SLC were co-cultured with T lymphocytes,and the immune response induced by dendritic cells was valued by assaying the interferon γ of the supernatant.Results:The dendritic cells that originated from CD34+ cells had typical dendritic stick and expressed CD1a,CD11c,CD40,CD80 and CD86.MTT method detected DCs-impulsed with Lenti-AFP/Lenti-SLC induced production as well as proliferation of CTL,this CTL specifically killed AFP positive HepG2 cells.CTL induced by Lenti-AFP/Lenti-SLC had stronger cytotoxic effect against HepG2 cells in comparison to unpulsed DC-primed T cell and HepG2 tumor lysate-primed T cell (P0.05 and P0.01).The highest anti-tumor activity was found with the ratio of effectiveness to the target was 50:1 (P0.01).Conclusion:DC-impulsed with Lenti-AFP/Lenti-SLC can induce production and proliferation of allogenic CTLs,which show antigen specific anti-tumor effect against AFP positive PHC cells.This finding lays the foundation ofimmunotherapy of PHC." @default.
