- W2371636470 abstract "Objective In this study we assessed the effects of calorie restriction (CR) mediated protection against chemotherapy damage on ovarian reserve. Methods Forty-eight female Sprague-Dawley rats were randomly divided into four groups: the NC group (fed ad libitum), the CR group (fed with 65% food intake of NC group),CTX group(injected with CTX and fed ad libitum) and CR+CTX group (injected with cyclophosphamide CTX and fed with 65% food intake of NC group), Ovarian reserve was examined by vaginal smears and follicle counting. The ovarian tissues were observed under microscope and the numbers of follicles at different developmental stages were calculated. Blood samples were collected and serous E2, LH, FSH levels were assayed by enzyme linked immunosorbent assay (ELISA). Results After the treatment with CTX, the activity and body weight of the rats were decreased, and the estrous cycle disappeared. The volumes of ovaries decreased. The necrosis of granulosa cells was observed in ovary by light microscope. The number of growing follicles significantly decreased and the number of atretic follicles significantly increased(P0.01). The CR group rats had a comparable number of primordial follicles compared to NC group. The CR+CTX group rats had significantly more primordial follicles and primary follicles than the CTX group. The serous levels of E2 ( P 0.01) were lower while the serum FSH and LH levels were significantly higher(P0.01) in the CTX group and CR+CTX group compared with the NC and the CR group. Conclusions These results suggest that CR may increase the ovarian follicular reserve and reduce the CTX-induced ovarian damage. CR protective effects may be due to its intervention in the transition from primordial to primary follicles and its reduction of oxidative stress." @default.
