- W2371724611 abstract "OBJECTIVE To determine the effect of carnosine on the expression and activity of astrocytic glutamate transporter-1(GLT-1)and glutamate aspartate transporter(GLAST)at a high extracellular glutamate level in primarily cultured cortical astrocytes.METHODS Astrocytes were identi-fied by immunofluorescent staining for glial fibrillary acid protein(GFAP)and then exposed to glutamate 10mmol·L-1 for 24,48and 72h,respectively.The cell death was measured by MTT assay and nucleus double staining.Astrocytes were pretreated with carnosine 5mmol·L-1 for 30min before glutamate 10mmol·L-1 was added to the cultures for 24h,and the changes in expression of GLT-1and GLAST proteins and mR-NAs was analyzed by immunohistochemistry and real-time PCR,respectively.Carnosine 5mmol·L-1 alone was applied to the cultures for 1-5dand the mRNA expression of GLT-1and GLAST was analyzed by real-time qPCR.High-performance liquid chromatography was used to determine the change in the extra-cellular glutamate level after astrocytes were pretreated with carnosine 5mmol·L-1 for 30min and then ex-posed to glutamate 1mmol·L-1 for 30min.RESULTS More than 95% of the cultured cells were astro-cytes as identified by immunofluorescent staining for GFAP.Glutamate 10mmol·L-1 insulted for 24and 48hdid not cause astrocytic cell death,whereas the insult sustained to 72hcaused decrease in cell viabili-ty to 82.0% of that of normal control and the apopotosis rate was increased to 24.7%.Glutamate expo-sure caused a slight but not significant increase in GLT-1and GLAST expression.Carnosine alone did not affect the mRNA expression of GLT-1and GLAST,whereas carnosine combined with glutamate enhanced GLT-1(P0.05),but not GLAST mRNA and protein expression.In addition,carnosine pre-treatment also significantly decreased the extracellular glutamate level.CONCLUSION Carnosine can promote a rapid increase in glutamate uptake and positive-regulate GLT-1but not GLAST expression in astrocytes exposed to high concentrations of glutamate,indicating that carnosine may be a potential anti-excitotoxici-ty agent in cerebral ischemia." @default.
