- W2372214576 abstract "Objective To investigate the mechanism of role of Th17 cells in pathogenesis of asthma in a murine model.Methods 30 mice were randomized into control group and asthma group.For the reproduction of a murine asthma model,the mice were first sensitized by 2mg OVA and 5mg aluminum hydroxide emulsion in 200μl of normal saline via intra-peritoneal injection on day 0 and day 14.From day 21 to 26,the mice were re-challenged with 1% OVA in saline aerosol in a total volume of 40ml for 40min per day.The control mice were given saline alone under the same conditions as the asthma group.The changes in cellular proportions in the BALF and the histological changes in lung tissue were observed.Th17 cells in the peripheral blood,thymus and lung tissue were detected by flow cytometry.Results The total cell number and the percentages of neutrophils,eosinophils and lymphocytes in BALF of the asthmatic mice were significantly higher than that in the control mice(P0.01).The neutrophil and eosinophil infiltration in pulmonary tissue was dramatically increased in asthmatic mice,but no such change was found in control mice.The percentages of Th17 cells in peripheral blood(3.62%±0.58%) and thymus(6.96%±1.35%) in asthmatic mice were significantly higher than that in control mice(0.68%±0.26% and 0.80%±0.30%,respectively,P0.01).While the number of Th17 cells in lung tissue did out show significant difference between asthmatic mice(0.64%±0.30%) and control mice(0.55%±0.23%,P0.05).Conclusions Th17 cells are probably involved in the pathogenesis of asthma.The cells may be released into blood from the thymus and then recruit neutrophils and eosinophils into airways,leading to inflammation of the airway in asthma." @default.
