- W2373184743 abstract "Objective To study the inhibitory effects of some local anesthetics on oxygen free radicals generation of polymorphonuclear neutrophils(PMNs) and its possible mechanisms.Methods Human venous blood was obtained from 12 healthy donors who had not used any medication for at least two weeks.Neutrophil suspension obtained with the purity exceeded 95% and the viability greater than 98% were devided into six groups:Group Ⅰand Ⅱ were pure reaction mixture and reference sample,respectively.Group Ⅲ～Ⅵ were severed as study groups which contented different local anesthetics.The cytochrome-c reduction assay was used to measure extracellular O_2~-production by actived PMNs and the reaction was actived by either fMLP or PMA.O_2~-dependent cytochrome-c reduction was determined by subtracting the reference value from the study sample value.Results At a concentration of 1×10~(-4)mol/L,ropivacaine,levobupivacaine,bupivacaine and tetracaine all could inhibited the O_2~-production of PMNs and their percents of the control response were(77.5±4.7)%,(75.8±6.4)%,(88.3±7.3)% and(61.3±5.1)% respectively(P0.01).Conclusions Local anesthetics inhibit the oxygen radicals production of activated PMNs;Stereoisomeric effects and lipid solubility may account for the different inhibitory effects of local anesthetics on PMNs." @default.
