- W2373702014 abstract "Objective:To study the follicular growth in organotypic culture supported by mesenchymal stem cell (MSC)-soft agar system.Methods:MSC were isolated,cultured,and further used as feeder cells. Cell surface markers of MSC were analyzed with fluorescence-activated cell sorter (FACS). The feeder cells were coated with 0.4% agar. Ovary slices from 4-week-old mice with thickness of 500 μm were imbedded in soft agar meanwhile spare agar were rejected.Vacant spaces between islets were fulfilled with conditioned medium (CM) of MSC containing 1.5 IU/ml PMSG.Half of the medium was substituted once every other day for estradiol and progesterone detection with RIAs.At day 6 of culture,the medium was replaced by CM supplemented with 1.5 IU/ml of hCG to induce final follicles maturation. Cumulus-oocyte complexes (COCs) were harvested under stereomicroscope 17h later. The competence of fertilization and embryo development were evaluated after in vitro fertilization (IVF).Results:All surviving follicles began to initiate growth at day 2. The follicles showed a significant increase in diameter with quantities and layers of granulosa cells multiplied. Follicles showed translucent areas within the granulosa cell mass around the oocyte after 17 h of hCG stimulus. The production of estradiol progressively increased during culture. The basal progesterone production stayed low in this organotypic culture which showed a dramatic increase after hCG stimulus. The percentage of fertilization in oocytes from organotypic culture was lower than those acquired from superovulation (62.5% vs 89%,P0.01) while no significant difference existed in blastocyst formation between them (82.4% vs 87.5%,P0.05).Conclusion:The MSC-soft agar system is benefit to follicular growth and maturation in organotypic culture.The mature oocytes from this system have good competence in fertilization and embryo development." @default.
