- W2374148399 abstract "【Objective】To explore the correlation between thymidylate synthease gene polymorphism and sensitivity to 5-Fu-based chemotherapy in gastrointestinal cancer.【Methods】PubMed,Institute for Scientific Information(ISI),EMBASE and China National Knowledge Infrastructure(CNKI) were searched for published case-control studies investigating the association between thymidylate synthease gene polymorphism and sensitivity to 5-Fu-based chemotherapy in gastrointestinal cancer.The odds ratio was calculated to evaluate the effectiveness of 5-Fu-based chemotherapy among three genotypes of gastrointestinal cancer patients.Fixed or random effect models were selected for odds ratio calculation.Publication bias was assessed.All statistical analysis was conducted with REVMAN 5.0 software.【Results】A total of 7 case-control studies involving 483 patients were analyzed in the study.Of all the 483 cases,the proportion of TS 5'-UTR 2R/2R,2R/3R genotypes and 3R/3R genotype were 57.8%(279/483) and 42.2%(203/483).The response rates to chemotherapy were 43.7%(122/279) and 34.8%(71/204).Compared the odds ratio and 95% confidence interval(CI) between the two groups was 1.87(0.81～4.28).In the population and tumor types subgroup analysis,the odds ratios for Asian,Caucasian populations,Stomach cancer,Colorectal cancer patients were 4.10(95% CI: 2.19～7.68),0.75(95% CI: 0.26～2.15),6.80(95% CI: 2.33～19.86),1.09(95% CI: 0.38～3.08) respectively.【Conclusions】TS gene 5`-UTR tandem repeat polymorphism is not associated with the sensitivity to 5-Fu-based chemotherapy in gastrointestinal cancer but in Asian patients and stomach cancer subgroups,the sensitivity of 2R/2R,2R/3R genotypes are higher than 3R/3R genotype." @default.
