- W2375762016 abstract "Objective To probe gene polymorphism and linkage disequilibrium analysis of -31 and -511 in the promoter of IL-1β in patients with inflammatory bowel disease, and thus to investigate the relationship between the gene polymorphism of IL-1β and inflammatory bowel disease. Methods Polymorphisms at -31 and -511 in the promoter of IL-1β were genotyped by PCR-RFLP method. Allelic frequencies were calculated, and linkage disequilibrium, accompanied by the disease-related haplotype, was analyzed in 48 patients with inflammatory bowel disease and 137 healthy controls. Results The proportions of genotypes of CC, CT and TT at -31 in the promoter of IL-1β in control healthy people were 22.63%, 56.93% and 20.44%, respectively, whereas those in patients with inflammatory bowel disease were 33.33%, 39.58% and 27.08%, respectively. There was no significant difference between the two groups (P0.05). The difference of allelic frequencies between the two groups was not of statistical significance either (P0.05). Similarly, the proportions of genotypes of TT, TC and CC at -511 in the promoter of IL-1βin healthy people were 21.17%, 52.55% and 26.28%,respectively, whereas those in patients with inflammatory bowel disease were 29.17%, 43.75% and 27.08%, respectively. There was also no significant difference between them (P0.05). The difference of allelic frequencies between the two groups was also not of statisticalsignificance.The association coefficient between IL1β-31C/T and -511T/C was 0.735 (D'= 0.915). The most predominant haplotype embracing the two loci was IL1β-31C/-511T(OR, 1.216; 95% CI, 0.763~1.937). Conclusion The proportions of genotypes of C/T and T/C are predominant at -31 and -511 in the promoter of IL-1β respectively in patients with inflammatory bowel disease. Gene polymorphism of IL-1β may be associated with particular ethnic groups. There is a strong linkage disequilibrium between IL1β-31C/T and -511T/C. IL1β-31C/-511T,the most predominant haplotype embracing the two loci, is strongly associated with IBD." @default.
