- W2376872471 abstract "Objective To develop a liquid chromatography-atmospheric pressure chemical ionization-tandem mass spectrometric method for the simultaneous determination of chlorpheniramine and caffeine in human plasma.Methods Chlorpheniramine,caffeine and internal standard were extracted from plasma with ethyl acetate and separated on a Diamonsil C18 column using a mobile phase of methanol-water-formic acid(V∶V∶V=80∶20∶0.5)at a flow rate of 0.5 mL·min-1.A tandem mass spectrometer equipped with atmospheric pressure chemical ionization source(APCI)was used as the detector and was operated in positive mode.Selected reaction monitoring(SRM)using the precursor-product ion transitions of m/z 275→m/z 230,m/z 195→m/z 138 and m/z 256→m/z 167 were performed to quantify chlorpheniramine,caffeine and internal standard(diphenhydramine),respectively.Results The assay was linear over the concentration range of 0.2-40.0 μg·L-1 for chlorpheniramine and 20.0~4 000 μg·L-1 for caffeine.The lower limits of quantification(LLOQ)were 0.2 μg·L-1 and 20.0 μg·L-1,respectively.The intra-day and inter-day precision was less than 13.4% and relative error was within±8.2%.Conclusions The developed method is simple,rapid,sensitive,specific and suitable for the simultaneous determination of chlorpheniramine and caffeine in human plasma." @default.
