- W2377156925 abstract "The present paper is aimed at introducing our research work on a biopolymer known as Poly(3-hydroxybutyrate-co-hydroxyvalerate)(PHBV),a bacterial derived biopolyester.As a nice candidate material used in tissue engineering,the application of PHBV should be promoted.Therefore,we have done an investigation on the cytobiocompatibility of PHBV material with 3-(4,5 dimethylthiazole-2-yl)-2,5-diphenyl tertrazolium bromide(MTT) assay in this paper.As an engineering practice,PHBV leaches used to be prepared first at normal(37 ℃) temperature for 72 hours and high(121 ℃) temperature for 1 hour in conformity with the Chinese national standard GB/T 16886.12.And PVC material was used for positive control as a rule.In order to determine the effects of PHBV on the cell growth,BHK-21 cells of mice were cultivated directly in the animal culture media(RPMI1640) containing either PHBV films or different concentrations of PHBV leaches at 37 ℃ and 5% CO_2 for one,three and five days.The optical density(OD) of BHK-21 cells was then measured and the relative growth rate(RGR) of cells was calculated based on the results of MTT assay.The cytobiocompatibility of PHBV was evaluated according to ISO10993 and GB/T16886.Our experimental results show that the toxicity grades of PHBV proves all zero,whereas the toxicity grades of PVC material used as the positive controls were as high as three or four.Thus,it can be seen that BHK-21 cell growth was not affected by PHBV material.The zero grade of PHBV leach at high temperature also indicated that it has a good chemical stability in a circumstance at high temperature.All the above results can prove that PHBV has a good cytobiocompatibility in vitro." @default.
