- W2378130573 abstract "Objective To study the clinical application of three kinds of flow cytometric methods for platelet counts. Methods Three kinds of flow cytometric methods for platelet counts by two-dimensional laser scatter method (AdVIA 120 method), RBC/platelet ratio method (RBC/PLT-R method) and absolute counting method by using fluorescent microbeads (TruCOUNT method) and respectively compared them with impedance method. Results Three kinds of flow cytometric methods correlated extremely well ( P0.001), at platelet in (1~646)×109/L, relative coefficient (r) 0.99. when platelet in(1~89)×109/L, r0.95. Comparision of TruCOUNT method with ADVIA 120 and RBC/PLT-R method, ADVIA120 was higher and RBC/PLT-R was lower. In low platelet counting, coefficient of variation of ADVIA120 was the smallest (3.46%), followed by TruCOUNT (5.2%), and the highest coefficient of variation was that of RBC/PLT-R (5.84%). Although impedance method and three kinds of flow cytometric methods for platelet counts show a correlation (r0.98, P0.001) too, platelet counts by impedance method were lower than that by three kinds of flow cytometric methods, and there was the abnormal error of platelet counting by impedance method in some cases. Conclusion It is proposed that three kinds of flow cytometric methods for platelet counts may be more accurate than impedance method. ADVIA120 automated blood cell analytic system is suitable for clinical routine in platelet counts, it was not only correlative well with immuoplatelet counting method of monoclonal antibody, but also simple and rapid. TruCOUNT method is an ideal method of platelet counting, which is accurate, sensitive and less influenced. So TruCOUNT method is applicable to counting low platelet and calibrating the blood cell analyzer. When the patients with severe thrombocytopenia need prophylactic platelet transfusion, TruCOUNT method can be used for counting platelets accurately and precisely." @default.
