- W2378761172 abstract "Objective To investigate the effect of Galactosylated human serum albumin 5-fluorouracil conjugate(Gal-HSA-5-FU) on proliferation and cell cycle in human hepatocarcinoma cells(Bel-7402).Methods The inhibitory rates of cell proliferation and IC50 values of Gal-HSA-5-FU were determined by MTT assay. Cell cycle of the tumor cells was analyzed with flow cytometry.Results Cell proliferation was inhibited after the cells were treated with various concentrations of Gal-HSA-5-FU(ranging from 0.953～30.500mg·L-1) for 24,48,and 72h,respectively. The inhibitory rates of cell proliferation were enhancing with the increase of drug concentration and treatment time(P0.01).The IC50 values of Gal-HSA-5-FU were(20.957±0.752),(11.793±0.546),and(5.843±0.735) mg·L-1 at 24,48 and 72h,respectively.The IC50 values at 48h and 72h were significantly lower than that at 24h(P 0.01).After the treatment of 24h,the proportion of Bel-7402 cells in the S phase increased,while the proportion in the G2/M phase decreased.There was little number of apoptotic cells according to the fluorescence staining assay.Conclusion Gal-HSA-5-FU can inhibit the proliferation of Bel-7402 cells in a dose-and time-dependent manner,and can arrest Bel-7402 cells in S phase." @default.
