- W2379595808 abstract "In this study,we aim to investigate the effects by nerve-growth-factor(NGF) and its receptor TrkA on the development of central cholinergic neurons and mRNA expression of the NGF and its receptor TrkA in hippocampus and parietal cortex.Wistar rats were divided into 3 groups:hypothyroid group(The young hypothyroid rats were constructed by rearing the mothers 0.02% methimazol water from 5 ~ 6 day of gestation to the day when they were sacrificed),hyperthyroid group(The young hyperthyroid were constructed by daily intraperitoneal injection of 0.4 μg/g body weight tetraiodothyronine after delivery),and normal control(The euthyroid rats were made by rearing the mother a normal diet and water).Cholinergic acetyltransferase-positive puncta was measured by immunohistochemistry,while NGF mRNA and TrkAmRNA expressions were measured by in site hybridization.As compared with agematched control(174.80 ± 5.10,171.40 ± 3.87,213.40 ± 16.93,274.60 ± 6.69),ChAT-positive puncta significantly decreased in parietal cortex and hippocampus of hypothyroid group(159.42 ± 5.26,162.37 ± 4.67,98.60 ± 7.67,101.50 ± 4.39) at postnatal 21 and 28 days(P 0.05 or P 0.01),but ChAT-positive puncta increased in same regions of hyperthyroid pups(312.50 ± 3.75,296.30 ± 6.13,324.60 ± 15.72,319.10 ± 8.85) at postnatal 21 and 28 days(P 0.01).The mRNA expression peak of NGF and TrkA in normal control appeared at P21 and P28 respectively,while ChAT-positive puncta peak was at P28.There was no peak in hyperthyroid rat.In hypothyroid team,the peak of mRNA expression of TrkA appeared at P28,ChAT-positive puncta peak at P21,and NGF peak at P28.As compared with age-matched euthyroid,the mRNA expressions of NGF and TrkA in parietal cortex and hippocampus was significantly lower in hypothyroid rats at P14,P21,P28(P 0.05 or P 0.01),but NGF mRNA expression was higher in hyperthyroid rats at P14,P21(P 0.05 or P 0.01),while TrkA mRNA expression showed no significant(P 0.05) in hyperthyroid pups.All these results indicate that the cholinergic neurons are very sensitive to hypothyroidism and hyperthyroidism in brain development;the change of mRNA expression of NGF and TrkA may affect central cholinergic neurons in brain development.It is conceivable that thyroid hormone effects upon ChAT could also have been mediated by NGF and its receptor TrkA." @default.
