- W2380051575 abstract "Objective:The objective of this study is to investigate the effect of alternating micro-current at intermediate frequency(100 kHz,20 mA) combined with chemotherapy agents for the treatment of human breast cancer cells MCF-7 in vitro.Chemotherapy agents include Epirubicin,Methotrexate,and Cisplatin.Methods:The MCF-7 cells in logarithmic growth phase were planted in 24-well plates,then divided into 4 groups randomly applied different treatment:control groups,electrical stimulation groups,chemotherapy groups,and combination therapy groups.The growth inhibiting rates to MCF-7 cells were determined by methyl thiazolyl tetrazolium(MTT) method.Apoptosis or death grates of MCF-7 cells were evaluated by Annexin V-FITC and PI double labeled flow cytometry.Laser scanning confocal microscopy was used to observe the intracellular content of Epirubicin and semi-quantitative analysis.Transmission electron microscopy was applied to examine the changes in morphology and structural of MCF-7 cells,in order to evaluate cells apoptosis.Results:Alternating micro-current at intermediate frequency used alone could inhibit the growth of MCF-7 tumor cells and the cell structures were in typical apoptosis states.Compared with cells chemotherapy groups,chemotherapy with alternating micro-current at intermediate frequency markedly inhibited the cell growth(P0.01).Also,the IC50 of chemotherapy drugs were decreased in the combination therapy groups.Alternating micro-current at intermediate frequency improved the content of Epirubicin(20 μg/ml) accessed to MCF-7 cells.Results of transmission electron microscopy showed that MCF-7 cells in combination therapy groups were damaged most seriously,including mitochondrial swelling,how foam body and lysosome.Conclusions:Alternating micro-current at intermediate frequency combined with chemotherapy could increase the inhibition rate of MCF-7 tumor cells in vitro,and it may be explored as a new breast cancer noninvasive modality.The possible mechanisms is that alternating micro-current at intermediate frequency can increase the concentration of chemotherapeutic agents accessed into cells and promote cells apoptosis." @default.
