- W2380093525 abstract "Objective:To explore the feasibility, loading parameters, and biological characteristics of intact human erythrocytes when they are used as a vincristine (VCR) delivery system.Methods: Human erythrocytes were loaded with VCR using a modified hypotonic pre-swelling technique.The loading parameters, including loaded amount, efficiency of entrapment, and erythrocyte recovery were evaluated through determination of intra-cellular and extra-cellular VCR amounts (HPLC) and physiological index of erythrocytes (auto-cytoanalyze). After storing at 4℃ for aspecified time, we measured the VCR amounts and observed hemolysis in the supernatant to compare the drug-releasing rate and storage stability of the carrier erythrocytes. Osmotic fragility was evaluated through exposing VCR-loaded erythrocytes to different hypotonic sodium buffers, and the morphology of VCR-loaded erythrocytes was observed with a phase contrast microscope. Results: The optimal amount of VCR loaded in 1×10~9 erythrocytes was obtained when the pre-swelling erythrocytes were mixed with 0.6% saline of VCR, with the loaded amount being 49.59 μg. When the erythrocytes were mixed with 0.6% saline of VCR for 20 min, the VCR-loaded amount became stable. The VCR-loaded amount of 1×10~9 erythrocytes was linearly correlated with VCR concentrations in the range of 50 to ~2 000 μg/ml(y=0.098 2 x-2.465 8,r=0.996 1).The efficiency of entrapment was (61.42±3.69)% and the percentage of erythrocyte recovery was (90.61±4.95)%. The osmotic fragility of VCR-loaded erythrocytes decreased, but there was no significant change in VCR-loaded erythrocyte morphology except that their sizes were slightly increased and were positively correlated with the amounts of VCR loaded in erythrocytes(r=0.988 0). The slow releasing rate of VCR accumulated for 4 h was ~(11.35±1.65)% and the carrier erythrocyte could be preserved for 5 d. Conclusion: These results suggest that,with good drug-loading parameters,human erythrocytes can be used clinically as a drug delivery system of VCR." @default.
