- W2380498123 abstract "The gonadotropin receptors, including luteinizing hormone receptor (LHR)and follicle-stimulating hormone receptor are essential for normal gonadal developmentand function by regulating steroidogenesis and cell proliferation. Although the sex ofbirds is controlled by sex chromosome, the differentiation of gonads is affected by manyfactors, for example, the sex steroid hormones which are synthesized by P450steroidogenic enzymes. Since the male and female gonads manifest differentmorphological patterns, their development might be modulated by differentsteroidogenesis and gonadotropic actions. In our experiment, the chicken gonads fromDay 10 of incubation to Day 14 posthatch were collected for studying the expression ofluteinizing LHR, P45017 a-hydroxylase (P450c17) and P450 aromatase (P450arom)mRNAs by Northern hybridization. The LHR mRNA transcripts of 3.0 and 1.5 kb weredisplayed and an additional 7.0 kb-transcript was detected only in posthatch gonads.During embryonic stage, the truncated 1.5 kb transcript was more abundant than that of3.0 kb, but less after hatching. In ovaries, the LHR mRNA level increased from Day 12of incubation and reached a peak two days later and then remained at a high level. Onthe contrary, the LHR mRNA levels in testes remained low during incubation andincreased immediately after hatching. The difference in the amount of LHR mRNAmight be related to sex differences in steroid synthesis. The changes of P450cl7 mRNA,both in the male and female, were very similar to those of LHR mRNA expression, inwhich a sex difference was also found. The P450arom mRNA manifested high expressionin ovaries, with a shift of the predominant transcript from 2.0 to 4.0 kb around hatching.The expression of the 2.0 kb transcript was significantly higher than that of the 4.0 kbtranscript during embryonic stage, but was lower after hatching. However, in testes, theP450arom mRNA transcript level remained much lower during embryonic and posthatchperiods examined. There was no significant difference in LHR and sterodogenic enzymemRNA levels between left and right testes at any corresponding days. Significantchanges of expression pattern of the three mRNAs might be accompanied by thedevelopment of the gonadal steroidogenic cells around hatching. The higher expressiono f LHR, P450c17 and P450 arom mRNAs in embryonic ovary indicates that thesteroidogenesis is more active in the female than in the male." @default.
