- W2382444470 abstract "Objective To explore the effect and mechanism of insulin on chemotherapeutic drug sensitivity of human lung adenocarcinoma cells A549. Methods The sensitivity of human adenocarcinoma cells A549 to DDP was observed by MTT methods, and IC30 values of the cells were calculated. The A549 cells were pretreated with insulin of various concentrations for different time period, then, cells sensitivity to DDP was evaluated with MTT assay. Flowcytometry was used to study the cell cycle distribution of the A549 cells of each group. The expressions of CyclinA and CyclinD1 were detected by Western Blot to analyze the chemosensitization mechanisms of insulin. Results The IC30 value of DDP was approximate to 36.87μg/ml. Insulin enhanced the inhibitory effect of DDP(36.87μg/ml) on cell proliferation when human lung adenocarcinoma cells A549 were treated with insulin(2.0～16.0mU/ml;8 h～16 h) as indicated by MTT assay(P0.01). Flowcytometry showed Insulin can increase the S phase arrest induced by DDP. Conclusion These datasuggest that pretreatment of insulin of appropriate concentration for the appropriate time could enhance the inhibitory effect of DDP on A549 cell growth. The mechanisms may be related to increasing the S phase arrest induced by DDP.The increased expression of cyclinA plays an important role in the S phase arrest induced by DDP." @default.
