- W2382806193 abstract "Aim The acute hypotensive effects and therapeutic effccts of iptakalim hydrochloride (Ipt) were evaluated compared with nifedipine,pinacidil,captopril and bisoprolol.Methods Blood pressure was measured with noninvasive method in conscious stroke prone spontaneously hypertensive rats (SHRsp).Results and Conclusion The systolic blood pressure were decreased by 19,23,31,49mmHg after oral administration of Ipt at the doses of 0.75,1.5,3.0,6.0mg·kg -1 ,respectively.The hypotensive effects were prominent at 3～5 hours after Ipt administration and persisted for 5～9 hours,the duration and intensity of Ipt were dose-dependent.Compared with nifedipine,pinacidil,captopril and bisoprolol,the hypotensive effects of Ipt was prominent and steady,rapid onset,long duration,lack of bradycardia and tachycardia.Chronic Ipt treatment at the oral dose of 1,2,4 and 8mg·kg -1 once a day for 30 days induced 23,25,32,37mmHg decrease of the systolic blood pressure in a dose-dependent manner,while had no prominent effects on heart rates.On d15 or d30 during chronic experiment,the hypotensive effect of Ipt was similar to those in acute experiment.No tolerance to the antihyertensive effect of Ipt on SHRsp was developed during 30d treatment." @default.
