- W2383182379 abstract "Objective To investigate the role of γδ T cells in acute Pseudomonas aeruginosa(P. aeruginosa)pulmonary infection during the early stage of the innate immune. Methods An acute P. aeruginosa pulmonary infection mice model was used. The wild infection group, IgG control group, IgG infection group, and anti-γδT-cell receptor(TCR)infection group were established respectively. Flow cytometry was used to detect the expression of interleukin(IL)-17-γδT cells in lung. After depletion of γδT cells by anti-γδTCR, the mRNA and protein levels of IL-17A and IL-17F were measured by quantitative real-time polymerase chain reaction(qRT-PCR) and enzyme-linked immunosorbent assay(ELISA). The bacterial load in lung and histological changes were assessed. Results 1 The expression of IL-17-γδT cells were obviously increased 8 h after infection with P. aeruginosa. 2 After infection with P. aeruginosa for 8 h, the mRNA and protein levels of IL-17A were significantly decreased in anti-γδTCR group when compared with IgG control group(P0.01), and that of IL-17F were not increased. Differential cell count of bronchoalveolar lavage fluid(BALF) in anti-γδTCR group showed an obvious decrease in neutrophils. 3 After infection with P. aeruginosa for 16 h, the anti-γδTCR mice group presented a significantly increased bacterial load in lung(174 folds) when compared with the IgG control mice, and displayed pathologically heavy perivascular inflammatory infiltration with the inflammation extending into parenchyma.Conclusions During the early stage of innate immune after acute P. aeruginosa pulmonary infection, γδ T cells are the primary source of IL-17A and play an effective role in the clearance of pathogens." @default.
