- W2384009471 abstract "Objective: To investigate the expression of P-gp protein and nuclear transcription factor κB in the brain of refractory epilepsy model,and explore whether topiramate(TPM) affects the express of P-gp and NF-κB in attempt to explore the role of P-gp and NF-κB resistance mechanisms of refractory epilepsy.Methods: Animal models of focal epilepsy were constructed by chemical kindling(injection of KA to hippocampus).All 84 Wister rats were randomly divided into three groups.N.S injection for control group(24),only KA injection for KA group(30) and KA injection with TPM for TPM group.TPM was given once every day for 14 days,and TPM was given two hours before KA injection at 14th day.After kindling,the epileptic rats were sacrificed at 6h,24h and 3d after KA injection,respectively.The expression of P-gp and NF-κB in the brain tissue was detected by immunohistochemistry.All data of different groups were analyzed.Results:①The P-gp expression in the hippocampus was increased significantly in the KA group compared with the control group(P0.01).The P-gp expression was increased 6 h after injection of KA(P0.01),and it was significant increased at 24h(P0.01),and it was descended 3d after KA injection(P0.01).There was no significant difference between the KA group and the TPM group(P0.05).②The NF-κB expression was increased significantly in the KA group compared with the control group(P0.01).The NF-κB expression was increased 6 h after injection of KA,and it was significant increased at 24h and 3d compared with that at 6h(P0.01),with no difference at 24h and 3d(P0.05).There was no significant difference between the KA group and the TPM group(P0.05).③The expression of P-gp and NF-κB in the hippocampus showed a positive correlation in KA group(r=0.86,P0.01).Conclusion: The expression of P-gp in the hippocampus is increased significantly in the animal models.P-gp may play an important role in the multidrug resistance of refractory epilepsy.TPM shows no increase in the P-gp expression,and the P-gp over-expression may be only the result of epilepsy paroxysm.A positive correlation between NF-κB and P-gp suggests both co-affects in the multidrug resistance of refractory epilepsy." @default.
