- W2384714567 abstract "Objective To observe the influence of toll-like receptor 4( TLR4) on expression of type 17 helper T cells( Th17 cells) related factors( IL-17A,IL-23 and RORrt) in the liver tissues of rats with ischemia-reperfusion injury,and to investigate the possible mechanisms. Methods Forty healthy male SD rats were divided into 4 groups,10 in each group: the sham operation group,model group,anti-TLR4 group and isotype antibody group. The carotid artery and femoral artery and vein of rats were separated in the sham operation group,and then intubated and injected with the same dose of heparin. Liver damage models of traumatic hemorrhagic shock reperfusion were made in the model group,anti-TLR4 group and isotype antibody group. The anti-TLR4 group and isotype antibody group were respectively injected with antiTLR4 antibody,isotype control antibody 50 μg from femoral vein 10 min before reperfusion,and the pace of injection was slow with microinjector. The rats were executed to obtain the tissues after treatment for 12 h. The TLR4 protein expression in each group was detected by Western blotting; the expression of IL-17A and IL-23 in liver tissues was detected by ELISA; IL-17A,IL-23 and RORrt mRNA changes were observed by RT-PCT. The correlation analysis on IL-17A,IL-23 and RORrt of the model group,anti-TLR4 group and isotype antibody group was conducted. Results The expression of TLR4 protein increased in the model group compared with that of the sham operation group( P 0. 01),and decreased after treatment with anti-TLR4 neutralizing antibodies( P 0. 01),and no significant difference was found in the isotype antibody group as compared with the model group( P 0. 05). Compared with the sham operation group,the expression of IL-17A and IL-23 was increased in the level of protein and mRNA,and RORrt mRNA was increased in the model,antiTLR4 and isotype antibody groups. Compared with the model group,IL-17A and IL-23 expression in the level of protein and mRNA,and the expression of RORrt mRNA was decreased in the anti-TLR4 group( P 0. 05 or P 0. 01). The level of IL-23 was positively correlated with the level of IL-17A,and IL-23 mRNA had a positive correlation with RORrt mRNA. Conclusions Neutralizing TLR4 can decrease the expression of IL-23,IL-17A and RORrt. The mechanism may be that blocking TLR4 can reduce IL-23 expression,and then affect the expression of RORrt which is the specific transcription factor of Th17,thus lead to the reduction of Th17 / IL17 expression in the liver tissues of rats with ischemia-reperfusion injury." @default.
