E-mail: g ujk@ mail. jlu. edu. cn Introduction Metformin ( 1, 1-dimethy lbiguanide) ( F ig. 1) is an or al ant i-hypergly cemic agent used in the t reatment o f non-insulin-dependent diabetes mellitus ( type II ) . Ow ing to it s w eight-decreasing and serum lipid-normalizing effects, it is especially recommended for obese pat ients [ 1, 2] . Various analyt ical methods have been described for the measurement o f metform in in biolog ical f luids, including gas chromatog raphy( GC) [ 3_ 5] , capil lar y elect rophor esis ( CE ) [ 6] and HPLC [ 7~16] with UV detection 7_ 12, 17] . HPLC-Mass spect rometry ( LC/ APCIM S/ M S ) of fers an at t ract ive alter nat ive to HPLC [ 18] . Due to the high polar ity of met fo rmin, the GC method requires a complex and is t ime-consuming 3_ 5] , w hile an ion-pair ex tr act ion w as used in the CE method to remove ionic substances from plasma samples [ 6] . T he repo rted HPLC method also sufferes f rom several disadvantages, such as lack of sensit iv ity [ 7_ 9] , using complex ext ract ion pr ocedures w hich need protein precipitat ion w ith evapo ration of the supernatant or tw o-step ex t raction including pro tein precipitat ion and liquid-liquid ext ract ion [ 10, 11] , or requiring the use of ult rafilt ration and column-sw itching systems [ 8] .
