- W2386185325 abstract "Objective To study the mechanism of β amyloid (Aβ) on neuronal apoptosis and the effect of Kidney tonifying Recipe (KTR). MethodsThe rat hippocampal neurons were treated with Aβ fragment 1～42 and the neuronal survival was assessed by double stained with fluorescein diacetate and propidium iodide. Apoptosis of neurons was detected with the use of Hoechst 33258 staining and TUNEL staining. The expression pf Caspase 3 was determined by using immunocytochemical SABC method. The transcription of mRNAs related to bcl 2, bax, c jun and c fos gene were observed by Northern Blot or RT PCR. Results In the Aβtreated neurons, viable cells decreased and condensation of chromatin and nuclear fragmentation were shown. The TUNEL staining revealed that the number of positive neuron increased. The expression of Caspase 3 elevated and the expression of bcl 2 mRNA decreased, but bax and c jun mRNA increased. However, transcription of c fos mRNA had no significant change. After the cells were treated with KTR, all the above indexes were improved. Conclusion The mechanism of Aβinduced neuron apoptosis was related to its regulation on the expression of apoptosis associated gene and the Caspase 3 pathway. KTR could attenuate the neurotoxic action of Aβand improve neuronal survival." @default.
