- W2386347256 abstract "Aim To detect the PAI-1mRNA expression induced by TGF-β1 in KFs using SYBR real time fluorescence quantitative RT-PCR method.Methods KFs were isolated from keloid tissue and cultured in vitro.The cells were stimulated with TGF-β1(1.25～20 pmol·L-1) for 3 h or TGF-β1(10 pmol·L-1) for 0.5~6 h.Then PAI-1 mRNA expression was detected by SYBR Green I real time fluorescence quantitative RT-PCR method.β-actin gene was used as the reference gene to calculate the relative expression of PAI-1 mRNA of each group.Results Compared with control group,TGF-β1(10,20 pmol·L-1) obviously induced PAI-1 mRNA expression in KFs,Whose expression ratio increased to 4.19 and 4.44 folds(P0.01) respectively; the PAI-1 mRNA expression ratio was up-regulated to 2.29 and 2.41 folds with TGF-β1(10 pmol·L-1) for 1,2 h(P0.05) and was up-regulated to 4.19 and 5.83 folds for 3,6 h(P0.01).These results suggested that the most optimal stimulation conditions of TGF-β1 were:10 pmol·L-1,3 h.Conclusion SYBR Green I real time fluorescence quantitative RT-PCR method can be adopted to detect PAI-1 mRNA expression induced by TGF-β1 in KFs.These results provide the foundation of further investigating the target gene regulation mechanism of TGF-β1 signal pathway in KFs and developing new drugs to treat keloid." @default.
