- W2386397722 abstract "Objective To investigate the effects of anisodamine on transient outward potassium current(Ito) in left ventricular myocytes in rabbits with acute myocardial infarction(AMI),and to explore the cellular(ionic) basis of anisodamine in treatment of antiarrhythmia.Methods 45 rabbits were randomly divided into three groups(15 in each group);acute myocardial infarction group(AMI group),anisodamine intervention group(Ani group) and sham operated group(SO group).Rabbits in Ani group were injected with anisodamine at a dose of 5 mg/kg via femoral vein 1 min before operation,while rabbits in AMI and SO group were injected with a similar dose of placebo(0.9%).Both AMI group and Ani group were rendered to AMI models by a ligation of the left anterior descending coronary artery,but SO group was subjected to the standard surgical procedures without coronary ligation.Single ventricular myocytes were isolated enzymatically from the epicardial zone of the infracted region derived from the hearts in the 3 groups and whole cell patch clamp technique was used to record Ito.Results IV curve of Ito in the 3 groups showed linear voltage dependence,and it was lower in AMI group when compared with that in SO goup and Ani group.When the voltage was at +60 mV,Ito current in AMI group,Ani group and SO group was[(10.56 ±1.95)pA/pF,n=12][(15.34 ± 2.55)pA/pF,n=12] and [(17.41±3.13)pA/pF,n=15] respectively,there were significant differences between AMI group and Ani group and so group(P0.01),but there was no sigilificant difference of Ani group and SO group(P0.05).Conclusion AMI can induce significant reduction in Ito,while pretreatment with anisodamine may attenuate this change,suggesting that anisodamine can reverse this electrical remodeling,which may be partly responsible for its antiarrhythmia effects." @default.
