- W2386534818 abstract "Objective To detect human telomerase RNA gene component(hTERC)gene amplification in cervical intra-epithelial neoplasia(CIN)and uterine cervix cancer,and to investigate the potentiality of hTERC gene copies as a marker of uterine cervix cancer early diagnosis.Methods The cervical specimens were collected from 95 gynecological outpatients in Southwest Hospital of Third Military Medical University.Five groups were divided based on the pathologic diagnosis,including normal group (n=10),cervicitis/CINⅠgroup(n=32),CINⅡgroup(n=20),CINⅢgroup(n=18)and uterine cervix cancer group(n=15).The thin-layer slides were made with physiologic saline method and TCT hypotonic method.The intracellular fluorescent signals of the cervical specimens(hTERC gene copies) were detected by using fluorescence in situ hybridization(FISH)with the dual-color probe(GLP TERC/CSP 3).The third chromosomal centromere probe was used as the inner-control;1 cervical cancer cell strain(Hela cells)and lymphocytes from normal bone marrow as positive controls;and normal cervical cells were applied as negative controls.Results The hTERC gene expression rate of uterine cervix cancer group(93.3%)and CIN group(65.79%)were significantly different from that of normal group(1.012%)and cervicitis/CINⅠgroup(3.13%)(P0.05).hTERC gene amplification was found in Hela cells and lymphocytes from normal bone narrow.The hTERC gene expression rate was associated with the severity of lesion precancerous.Conclusion hTERC gene involves in the occurrence and development of cervical intra-epithelial neoplasia(CIN)and uterine cervix cancer.As a marker of CIN and uterine cervix cancer hTERC gene's detection may contribute to early diagnosis of uterine cervix cancer and treatment monitoring of cervical intra-epithelial neoplasia." @default.
